Description: Pyronin Y is a fluorescent probe which stains double stranded RNA in living or fixed cells as well as in tissues. Pyronin Y forms fluorescent complexes with double-stranded nucleic acids, especially RNA, enabling semi-quantitative analysis of cellular RNA in flow cytometry. When used in living cell preparations, it is commonly combined with 50-100µM verapamil to prevent efflux of the dye. It has been used to ascertain the cell cycle state of stem cells. Pyronin Y is widely used in combination with methyl green to selectively and differentially stain nucleic acids following polyacrylamide gel electrophoresis. In a dichromatic dye solution, Pyronin Y stains RNA red, while methyl green stains DNA green. It has also been shown to accumulate in mitochondria of viable cells. Spectral data: λex 546nm; λem 565nm (in DMSO).
